Reagents
Sent at Room Temperature
Tube rack containing 5 aliquots of the following reagents
- STET buffer 1-2 ml each
- SOC 2 ml each
- Isopropanol 1-2 ml each
- 70% ethanol 1-2 ml each
- sterile water 1-2 ml each
PLUS
- 1 bottle of 10X TBE (200 ml, includes spare)
- 2 wide mouth jars with small amounts of 95% ethanol (for sterilizing cell spreaders), with MSDS labels on jars
- 2 microwaveable tubs with 0.8g of Agarose in each (plus 1 tub for each additional gel required)
- 2 tubes loading dye (1 - 1.5 ml each)
- 1 tube (24 ul) Sybr safe DNA gel stain ( for 2 gels includes spare)
- 12 MacConkey-Amp plates (10 for class plus 2 spare)
- 3 Agarose-Kan plates (teacher's use only)
Sent on Wet Ice (crushed ice in Ziplock bags)
Aliquots below all include spare amounts to compensate for minor spillage etc.
- 12 tubes E.coli pellets
- RNase (1 tube 25 ul)
- PCR rxn mix (1 tube 100 ul)
- PCR template plus primer (1 tube 100 ul)
- 2X ligation butter (1 tube 60 ul)
- T-vector (aka T-Easy) from ligation kit (6 ul diluted with 6 ul water to make up 12 ul)
- T4 ligase (1 tube 12 ul)
- 10X restriction buffer (comes with EcoR1 when we buy it) (1 tube 30 ul)
- DNA marker (aka Ladder) (1 tube 15 ul)

These are quantities PER CLASS and includes extras
- competent cells (12 tubes)
- lysozyme (1 tube 0.01 gm)
- EcoR1 enzyme (1 tube 12 ul) for digest
